The sericulture industry plays a very important role in our national economy. Silkworm (Bombyx mori) is always regarded as a model animal and biological reactor. There have been detailed studies on the structure, expression and control and molecular evolution of silk genes. However, few, if any, reports are available on the localization of structural genes in silkworm by molecular cytogenetics. The present experiment has tentatively localized theFib-H gene at the distal end of the 25th linkage group, namely at the 25-0.0 position, and verified thatFib-H has only one locus, thus providing a temporary solution to the problem about its localization.  相似文献

应用基因组原位杂交技术(GISH)对普通小麦(Triticum aestivumL.)和长穗偃麦草[Agropyron elongatum(Host)Beauv,2n=10x=70]杂交后选育出的蓝粒小麦蓝-58及其诱变后代的染色体组成进行了鉴定。结果表明,GISH可方便地检测到小麦遗传背景中的长穗偃麦草染色体或易位的片段。如前人报道,蓝-58(2n=42)是一个具有2条长穗偃麦草4E染色体的异代换系(4E/4D)。LW004可能是一个具有两对相互易位染色体的纯合系,其田间表现磷高效特性,LW43-3-4为41条染色体的蓝单体(40W 1’4E),种子颜色为浅蓝色,通过此法还检测出一些染色体结构发生很大变异的材料如4E的单端体(40W 1‘4E),种子颜色为浅蓝色,通过此法还检测出一些染色结构发生很大变异的材料如4E的单端体(40W 1‘t4E)以及组型为39W 1‘4E 1‘t4E的个体,此项研究结果更为直观地表明控制蓝粒体状的基因的确在来自长穗偃麦草的染色体上。同时说明有效的突变方法与灵活方便的检测手段的有机结合在染色体工程材料的创制和染色体工程育种中起着至关重要的作用。  相似文献

电子克隆是基因克隆的新策略,以小麦胞质葡萄糖-6-磷酸脱氢酶cDNA(Tagpdl克隆)序列为信息探针,在GenBank水稻nr数据库中找到高度同源的水稻基因组序列,通过人工序列拼接及RT-PCR确认得到了水稻该基因的全长cDNA序列,命名为OsG6PDH,OsG6PDH与小麦Tagpdl克隆的DNA一致率为88%,推导的氨基酸序列与小麦,番茄,烟草的胞质葡萄糖-6-磷酸脱氢酶基因的一致率分别为89%,79%,80%,经RT-PCR表达谱分析,OsG6PDH在水稻幼穗,胚,根,叶中都有表达,在幼穗与根中表达略高,另外,讨论了利用水稻基因组信息的电子克隆方法克隆水稻功能基因的可行性。  相似文献

The landscapes of the French island of Corsica are the result of thegrazing tradition which has always occupied a pivotal position in the island'seconomy. Furthermore, in the last fifty years, a reduction in the pressure fromhuman activity has lead to changes in plant cover. This article intends tostudythe evolution of dynamic mechanisms during the secondary successions and theeffect of changes in vegetation on plant diversity. Our study was based on asimultaneous study of 375 plots carried out between May and September from 1996to 1998. We studied the floristic composition of each stage both qualitatively(life form, adaptive strategy and seed dispersal mode) and quantitatively(species richness, equitability, Shannon index). The results show a change inspecies and environmental conditions, with a progressive move from heliophilicgrassland and low shrub species with an S or SR strategy, anemochoric and withahigh tolerance for soil quality to a final stage of dense forest composed ofcompetitive (C or CS) zoochoric or barochoric phanerophytes. Moreover, theturning point in Corsica in terms of floristic diversity lies between 20 and 55years after land abandonment. Successions are due to a series of rapidinteractions. Thus, when a site is given reserve or protected area status, weneed to take into account the fact that it will evolve according to thebiological traits of the species. From a research point of view, we need tolookat process diversity and the role of each of the species present during thesuccessions.  相似文献

Physical processes determine to great extent the habitat of hydrobionts, as well as the transfer and sedimentation of substances, the intensity of pollution and rate of natural purification of water bodies. Mathematical models of different levels of complexity, developed to investigate hydrophysical processes in lakes, are discussed in this paper. The numerical algorithms and computer programmes described can be used to determine the influence of the morphometric characteristics and weather on the temperature regime of Lake Shira (Khakasia, Siberia) and the pattern of the wind currents. Examples of calculating the temperature regime in the context of a one- dimensional model and of calculating the parameters of wind currents in the water body of a simple geometrical form are given.The study suggests that the pattern of wind currents in Lake Shira is significantly affected by the density stratification, which depends not only on temperature but also on salinity. In order to construct a realistic pattern of currents a 3-D computer model of Lake Shira must be developed and used to estimate the validity of the two-dimensional and one-dimensional models. The present research can be further developed by extending the obtained algorithms to three-dimensional problems, taking into account the heat exchange, salinity and the geometry (bathymetric contours) of the water body. Calculations can be made for Lake Shira and the obtained data used in biophysical models.  相似文献

The effect of five antibiotics: carbenicillin, chloramphenicol, cefotaxime, kanamycin and hygromycin on the organogenesis from callus cultures of Coryphantha elphantidens (Lem.) Lem. have been studied. Carbenicillin and cefotaxime stimulated shoot regeneration from callus. All antibiotics under study suppressed rooting of in vitro formed shoots. After five sequential subcultures on kanamycin supplemented medium, antibiotic resistant callus was obtained. To study the impact of kanamycin on resistant callus, total protein content was also studied. Selected callus showed a remarkable increase in callus mass. Antibiotic resistant plants have been selected by screening callus pieces on kanamycin supplemented media. Total protein content increased with subsequent subcultures in kanamycin resistant callus. The kanamycin selected shoots withstood the stability test after 2 months on antibiotic free medium. Plants were raised from the callus, which formed roots in 20 mg dm–3 kanamycin, which was under study.  相似文献

Sago, a processed (gelatinized) edible starch, was successfully used as a gelling agent in culture medium. The efficacy of sago-gelled (80 g dm–3) medium was studied in ten potato (Solanum tuberosum L.) genotypes during micropropagation and minimal growth conservation. Sago starch provided a firm gelling surface throughout the entire culture period, and fostered optimum plantlet growth in terms of shoot height, number of nodes per plant, number of leaves and fresh mass. No softening of the sago-gelled medium occurred over prolonged (six months) storage. The study showed that sago starch could be used as a substitute to agar in culture medium to substantially reduce the medium cost.  相似文献

We have analyzed the phylogenetic and genomic relationships in the genus Setaria Beauv. including diploid and tetraploid species, by means of the molecular diversity of the 5S rDNA spacer and chromosomal organization of the 5S and 18S-5.8S-25S rDNA genes. PCR amplification of the 5S rDNA sequences gave specific patterns. All the species studied here share a common band of about 340 bp. An additional band of an approximately 300-bp repeat unit was found for Setaria verticillata and the Chinese accessions of Setaria italica and Setaria viridis. An additional band of 450 bp was found in the sole species Setaria faberii. Fluorescent in situ hybridization was used for physical mapping of the 5S and 18S-5.8S-25S rDNA genes and showed that they are localized at two separate loci with no polymorphism of chromosome location among species. Two chromosome pairs carrying the 5S and 18S-5.8S-25S rDNA clusters can now be unambiguously identified using FISH. Phylogenetic trees based on the variation of the amplified 5S rDNA sequences showed a clear separation into four groups. The clustering was dependent on the genomic composition (genome A versus genome B) and confirmed the closest relationship of S. italica and S. viridis accessions from the same geographical region. Our results confirm previous hypotheses on the domestication centers of S. italica. They also show the wide difference between the A and B genomes, and even clarify the taxonomic position of S. verticillata. Received: 28 August 2000 / Accepted: 27 January 2001  相似文献

The aim of the study was to characterize genomic relationships among cultivated tomato (Lycopersicon esculentum Mill.) (2n=2x=24) and diploid (2n=2x=24) non-tuberous wild Solanum species (S. etuberosum Lindl.). Using genomic in situ hybridization (GISH) of mitotic and meiotic chromosomes, we analyzed intergeneric somatic hybrids between tomato and S. etuberosum. Of the five somatic hybrids, two plants were amphidiploids (2n=4x=48) mostly forming intragenomic bivalents in their microsporocytes, with a very low frequency of multivalents involving the chromosomes of tomato and S. etuberosum (less than 0.2 per meiocyte). Tomato chromosomes showed preferential elimination during subsequent meiotic divisions of the amphidiploids. Transmission of the parental chromosomes into microspores was also evaluated by GISH analysis of androgenic plants produced by direct embryogenesis from the amphidiploid somatic hybrids. Of the four androgenic regenerants, three were diploids (2n=2x=24 or 2n=2x+1=25) derived from reduced male gametes of the somatic hybrids, and one plant was a hypertetraploid (2n=4x+4=52). GISH revealed that each anther-derived plant had a unique chromosome composition. The prospects for introgression of desirable traits from S. etuberosum into the gene pool of cultivated tomato are discussed. Received: 2 August 2000 / Accepted: 4 December 2000  相似文献

2003年7月和2004年1月对海南省黎母山自然保护区两栖动物资源进行了调查,结果表明,该区内有两栖动物1目5科13属25种,全部为东洋界种类,其中华南区成分有15种,华中华南区有10种,分别占黎母山两栖动物总数的60%和40%,海南特有种有7种,具有典型的华南区系成分和热带山地林灌动物群特征。国家Ⅱ级保护动物有虎纹蛙1种;数量优势种有黑眶蟾蜍、小湍蛙、泽蛙、细刺蛙、海南溪树蛙和背条跳树蛙;保护区内两栖动物物种多样性指数为37109,均匀性指数为08321。  相似文献
